Development, validation and application of a rapid and sensitive method for determining trigonelline and caffeine by UHPLC-DAD present in roasted and ground coffee

Resumo: Trigonelline and caffeine are alkaloids from the group of xanthines of great biological importance. They are found in numerous foods and drugs, acting in different ways, with stimulating action, being precursors of bioactive compounds and, as prodrugs. They arouse growing interest in their contents determination by the diversity of applications, being used, for example, as markers in the differentiation between arabica coffee (Coffea arabica) and robusta (Coffea canephora). For this reason, this study proposed the development of a sensitive chromatographic method for the rapid determination of trigonelline and caffeine in coffee by ultra high-performance liquid chromatography with diode array detection (UHPLC-DAD). The developed method was validated according to ANVISA RE 899/2003 and applied in the analysis of commercial roasted and ground coffee. The method was suitable for all merit figures, with a linear working range of 0.20 to 2.00 %(w/w) for trigonelline and from 0.60 to 6.00 %(w/w) for caffeine, with limits of quantification of 0.03 and 0.06 %(w/w) and detection of 0.01 and 0.02%(w/w), respectively. It presented precision as repeatability with relative standard deviation (RSD) of 0.12% for trigonelline and 0.55% for caffeine, with RSD for the intermediate precision of 0.52 and 1.14%, respectively, with average accuracy of 99.80% in 3 levels of low, medium and high concentration, for both compounds. The method intends to assist in the quality control of commercial roasted and ground coffee.
