Analysis of statins in human plasma by gas chromatography-mass spectrometry using SPME and in situ derivatization sample preparation

Resumo: Therapeutic drug monitoring allows the individualization of dosing regimen, ensuring the clinical efficacy and minimizing adverse effects caused by the prescribed drugs. Statins belong to the group of inhibitors of the enzyme 3-hydroxy-3-methylglutarylcoenzyme A redutase (HMG-CoA). They induce a significant reduction in total cholesterol, low density lipoprotein and plasma triglycerides, therefore they are used in the treatment of hypercholesterolemia. Currently, statins have been monitored because they present undesirable side effects. This paper, describes analysis of fluvastatin, simvastatin and atorvastatin by solid phase microextraction and gas chromatography coupled to mass spectrometry (SPME-GC/MS) in human plasma for therapeutic drug monitoring. The conditions of extraction and derivatization were optimized using experimental design and evaluating the influence of the main parameters. The method presents linearity in the range of 20-500 ng mL-1 with precision lower than 14% and limit of quantification of 20 ng mL-1 and 50 ng mL-1 for fluvastatin and simvastatin, respectively. The proposed methodology was applied to samples of plasma from patients under simvastatin treatment.
